Making Nutrient Agar

Agar recipe & procedure from "sciencestuff.com"

 

 

 

 

 

 

 

 

 

  • Determine agar mass required according to information found in website above.
  • Add agar to dry beaker (try to keep it off sides)
  • Add water to beaker.
  • Microwave solution in 30 second intervals, swirling each time (but try not to get solution too far up the side of beaker!)
  • Once agar is dissolved, pour into petri dishes using aseptic technique.

 

 

 

 

 

 

 

 

  • Pour enough agar into dish to completely cover the bottom of the Petri dish and cover immediately.
  • After agar has set (solidified), turn dishes upside down so that they sit on their lids. Label bottom of dishes with appropriate information (name, experiment, recipe, etc.)

 

 

 

 

  • Store in the refrigerator if unused, or store in 32-37o incubator if you want to promote growth of bacteria.
